A 24‐Week, Phase IIa, Randomized, Double‐Blind, Placebo‐Controlled Study of Ziritaxestat in Early Diffuse Cutaneous Systemic Sclerosis

This study aimed to investigate the effectiveness, safety, and tolerability of ziritaxestat, a selective autotaxin inhibitor, in patients with early diffuse cutaneous systemic sclerosis (dcSSc). The 24-week study involved adults with dcSSc who were given either ziritaxestat or a placebo. The primary measure of efficacy was the change in modified Rodnan skin score (MRSS) after 24 weeks. The results showed that patients receiving ziritaxestat had a significantly greater reduction in MRSS compared to those on placebo. Ziritaxestat was well-tolerated, with headache and diarrhea being the most common side effects. Biomarker analysis indicated that ziritaxestat may reduce fibrosis. Overall, ziritaxestat shows potential for improving skin involvement in patients with dcSSc.
