A narrative review of the role of Eph receptors in head and neck squamous cell carcinoma

The present review focuses on the role of Eph receptors, a type of tyrosine kinase receptor, in the progression of head and neck squamous cell carcinoma (HNSCC) and their potential as therapeutic targets. The researchers conducted a comprehensive search of electronic databases to identify relevant studies up until August 2022. Among the Eph receptors, EphA2 and EphB4, along with ephrin-B2, were extensively studied in HNSCC. Overexpression of EphB4 and ephrin-B2 consistently correlated with poor outcomes, suggesting their potential as prognostic markers. High expression of EphA3 and EphB4 was associated with radioresistance in HNSCC. Clinical trials are currently underway to investigate the efficacy of blocking EphB4-ephrin-B2 in combination with standard treatment for HNSCC. However, further research is needed to understand the biological role and complexity of this family of tyrosine kinase receptors in HNSCC, with a focus on avoiding heterogeneity among different subsites of the disease.
