A novel integrated approach to predicting cancer immunotherapy efficacy

A new study has characterized the neoantigen landscape in lung cancer and melanoma and developed a composite score called NEO2IS that improved the prediction accuracy of patient responses to immune-checkpoint blockades (ICBs). The study found that T-cell receptor repertoire diversity was consistent with the neoantigen heterogeneity under evolutionary selections. The study also defined a neoantigen intratumor heterogeneity score (NEOITHS) that reflected the infiltration degree of CD8+ T lymphocytes with different differentiation states and the impact of negative selection pressure on CD8+ T-cell lineage heterogeneity or tumor ecosystem plasticity. The study helps enhance the understanding of evolving tumor-immune interplays and improves the prediction of ICBs efficacy.
