
The study identified the cellular changes in preterminal and terminal bronchioles (pre-TBs/TBs) in chronic obstructive pulmonary disease (COPD) and determined their cellular origin. The researchers used a novel method of distal airway dissection and performed single-cell transcriptomic profiling of cells from healthy lung donors and COPD patients. They identified region-specific cellular states and unique terminal airway-enriched secretory cells (TASCs) found only in distal airways lost in COPD pre-TBs/TBs. The researchers also found that basal cells residing in pre-TBs/TBs were the cellular origin of TASCs and that regeneration of TASCs was suppressed by interferon-gamma (IFN-γ). These findings suggest that altered maintenance of the unique cellular organization of pre-TBs/TBs represents the cellular basis of distal airway remodeling in COPD.
