AdV-CIITA Gene Therapy Boosts Immunity Against Glioblastoma

In a study using adenovirus (AdV) armed with the CIITA gene, surface MHC-II expression was induced in glioblastoma (GB) cell lines and organoids, promoting immune-mediated tumor cell death. Both wild-type and mutant Ad-CIITA triggered T-cell responses, suggesting an MHC-II-independent mechanism. Tumor cell killing required CD8+ or CD4+ T-cells and direct contact with GB cells. No canonical T-cell-mediated death pathways were activated. These findings highlight the potential of AdV-mediated CIITA gene therapy to enhance T-cell immunity, offering a new approach for combating the immunosuppressive glioblastoma microenvironment.
