ANGPTL3 Deficiency and Risk of Hepatic Steatosis

This study suggests that targeting ANGPTL3 (angiopoietin-like 3 protein) to reduce triglycerides and LDL cholesterol levels may not lead to hepatic fat increase, as previously observed with vupanorsen treatment. The study recruited individuals with ANGPTL3 deficiency due to loss-of-function mutations, finding lower cholesterol and triglyceride levels but no significant difference in hepatic fat or steatosis. Mendelian randomization analysis also indicated no causal relationship between ANGPTL3 and hepatic fat. This study challenges concerns about hepatic fat associated with ANGPTL3 inhibition, offering hope for safer therapeutic approaches.
