
In a hollow-fiber model using THP-1 cells infected with M. avium, bedaquiline added to azithromycin, ethambutol, and clofazimine showed no additional killing effect. Both regimens achieved similar intracellular and extracellular kill rates, and macrolide-tolerant subpopulations emerged regardless. The findings question the utility of bedaquiline as a second-line agent for MAC pulmonary disease treatment.
