This study aimed to investigate the relationship between immune cell populations, gene expression in abdominal subcutaneous adipose tissue (aSAT), and circulating inflammatory markers with liver and skeletal muscle insulin sensitivity in individuals with overweight and obesity. The study found that the relative abundances of circulating T cells, nonclassical monocytes, and CD56dim CD16+ natural killer cells were inversely associated with liver insulin sensitivity. The inverse association between circulating (classical) monocytes and liver insulin sensitivity was confirmed in a replication cohort. However, immune cell populations in aSAT were not related to insulin sensitivity. Gene expression of interleukin 6 and CD14 in aSAT was positively associated with muscle insulin sensitivity but not liver insulin sensitivity. Overall, the study suggests that immune cell populations and inflammatory gene expression in aSAT show distinct associations with liver and muscle insulin sensitivity.
