Collagen VII, a key extracellular matrix protein, forms anchoring fibrils (AFs) that enhance tissue integrity in stratified squamous epithelial tissues. AFs anchor the basement membrane to the dermis by looping around interstitial collagen fibrils. Initially, it was unclear whether collagen VII originated from mesenchymal or epithelial sources. Recent studies using species-specific antibodies and mouse genetic models revealed that collagen VII is derived from epidermal keratinocytes and dermal fibroblasts. This dual origin supports normal basement membrane turnover and postnatal growth. These findings offer insights into collagen VII's role in homeostasis, development, and disease, influencing molecular corrective therapies.
