Colorectal Cancer Metastasis: How MicroRNA 483-3p Affects NDRG1 Downregulation and ERBB3/AKT Axis Activation

The article discusses a study conducted on metastatic colorectal cancer and stem-like cell cultures (m-colospheres) which shows that overexpression of microRNA 483-3p (miRNA-483-3p) enhances tumor aggressiveness. Transcriptomic analyses and functional validation revealed that miRNA-483-3p directly targets NDRG1, a metastasis suppressor involved in EGFR family downregulation, and activates the ERBB3 signaling pathway leading to the activation of transcription factors regulating epithelial-mesenchymal transition (EMT). The study found that miRNA-483-3p expression inversely correlated with NDRG1 and directly correlated with EMT transcription factor expression and poor prognosis in human colorectal tumors. The results suggest that miRNA-483-3p, NDRG1, and ERBB3-AKT signaling are linked and can support colorectal cancer invasion, which can be targeted therapeutically with selective anti-ERBB3 antibodies.
