Copy Number Alterations Predict Development of OSCC from Oral Leukoplakia

Oral leukoplakia (OLK) is a potentially malignant disorder, and predicting its transformation into oral squamous cell carcinoma (OSCC) is crucial for effective management. This study analyzed 431 OLK samples with long-term follow-up, assessing genomic copy number alterations (CNAs) as potential biomarkers. CNA events increased with dysplasia severity, and OLK samples progressing to OSCC exhibited more CNAs. A CNA score model accurately predicted malignant transformation risk, outperforming histopathological grading. Subgroup analysis showed the CNA score's ability to predict transformation across different dysplasia severities. The CNA score emerges as a promising biomarker for predicting OLK malignancy, aiding in personalized management.
