Correlation between FDG-PET-CT and T cell profiling in predicting response to PD-1 blockade in NSCLC

The study explored the use of early response evaluation techniques to identify biomarkers for PD-(L)1 checkpoint blockade in non-small cell lung cancer (NSCLC). The study found that increased total lesion glycolysis (TLG) had a strong reverse correlation with overall survival (OS). Responders had decreased progressors and increased Treg frequencies on-treatment. Detectable PD-1-expressing CD8+ T cell frequencies decreased in responders but remained stable in progressors. Changes in activated Treg rates in TDLN were strongly correlated with ΔTLG and reversely correlated with OS. The study highlights the need for better biomarkers for PD-(L)1 checkpoint blockade in NSCLC.
