Decitabine Degrades KMT2A and Synergizes with Menin Inhibition in Leukemia

Decitabine (DEC), a hypomethylating agent, not only targets DNA methyltransferase DNMT1 but also degrades the leukemic driver KMT2A, showing potential for enhancing leukemia treatment. This study demonstrates that DEC reduces KMT2A protein expression, leading to cell cycle disruption and decreased leukemia cell proliferation. Furthermore, DEC synergizes with the menin inhibitor revumenib (REV) to enhance therapeutic effects. These findings highlight a novel mechanism for DEC in adult acute lymphoblastic leukemia (ALL) and suggest its potential as an effective treatment option in combination with menin inhibition.
