Dinaciclib Boosts Cancer Cell Radiosensitivity

Dinaciclib, a pan-CDK inhibitor, has potential as an adjuvant therapy with radiotherapy in cancer treatment. This study shows that dinaciclib enhances radiosensitivity in lung and colon cancer models (A549, HCT 116) but not in others (H1299, HT-29). Unlike other CDK inhibitors, dinaciclib did not affect ATM signalling or cell cycle changes post-radiation but significantly increased apoptosis. The mechanism involves CDK12 inhibition, leading to decreased BRCA1 expression, reduced homologous recombination (HR), and promotion of nonhomologous end joining (NHEJ), resulting in ionising-radiation-induced cellular senescence in a TP53-dependent manner.
