
The study links extracellular matrix (ECM) stiffening to cancer stem cell (CSC) activation in oral squamous cell carcinoma. ECM stiffness induces CD44+ stem-like traits through ITGB1-FERMT1-mediated mechanotransduction, which activates Wnt signaling via CK1α degradation. Targeting ECM stiffness and mechanotransduction reduces CSC-driven chemoresistance in vivo, identifying the ECM/ITGB1/FERMT1/Wnt axis as a potential therapeutic target.
