
A systematic review and meta-analysis involving 23 studies and 6,879 patients across 16 countries has shed light on the effectiveness of primaquine in preventing Plasmodium vivax recurrence. The research found that a high total dose of primaquine (7 mg/kg) significantly reduced P vivax recurrences compared to no treatment or a low total dose (3.5 mg/kg). The risk of recurrence at day 180 was 8.1% with high-dose primaquine, 19.3% with low-dose, and 51% without treatment. The study suggests that increasing primaquine dosage can cut recurrences by over 50% in most endemic areas, with manageable gastrointestinal side effects.
