Effectiveness and Safety of XEN1101 in Adults with Focal Epilepsy: Phase 2b Trial

A study assessed the potential of XEN1101, a novel Kv7.2/Kv7.3 potassium channel opener, in managing focal-onset seizures (FOSs). This phase 2b trial, conducted from January 30, 2019, to September 2, 2021, involved adults experiencing ≥4 monthly FOSs while on stable ASM treatment. The trial, spanning 97 sites in North America and Europe, randomly assigned patients to XEN1101 (25, 20, or 10 mg) or placebo for 8 weeks. The primary endpoint was the percent change in monthly FOS frequency, showing a significant dose-dependent reduction with XEN1101. The treatment was well-tolerated, with no reported fatalities, supporting further clinical development.
