Efficacy and Safety of Brodalumab, an Anti-interleukin-17 Receptor A Monoclonal Antibody, for Palmoplantar Pustulosis

This phase 3, randomized, double-blind, placebo-controlled trial assessed brodalumab's efficacy and safety in Japanese patients with palmoplantar pustulosis (PPP). Participants received brodalumab 210 mg or placebo subcutaneously every 2 weeks for 16 weeks. Brodalumab significantly reduced the Palmoplantar Pustulosis Area Severity Index (PPPASI) score compared to placebo, with notable improvements in secondary endpoints like PPPASI-50/75/90 response rates and Physician’s Global Assessment scores. Adverse events, primarily mild infections, were more frequent with brodalumab. The study supports brodalumab as an effective treatment option for moderate to severe PPP in Japanese patients.
