ENGOT-en11/GOG-3053/KEYNOTE-B21: A Randomised, Phase 3 Study of Pembrolizumab in High-Risk Endometrial Cancer

In a study evaluating adjuvant pembrolizumab combined with chemotherapy for high-risk endometrial cancer (EC) patients with no residual disease post-surgery, no overall improvement in disease-free survival (DFS) was observed compared to placebo. Among the 1,095 participants, 2-year DFS rates were similar between the pembrolizumab and placebo groups. However, in the dMMR subgroup, pembrolizumab showed a significant DFS benefit (HR 0.31), while no such benefit was seen in the pMMR subgroup (HR 1.20). Safety profiles were acceptable with no grade 5 adverse events.
