A study aimed to investigate the mechanism of fibrocartilage repair after microfracture, a common treatment for cartilage defects. This study used single-cell transcriptional assays to identify cell subsets and specific marker genes at different repair stages after microfracture in Bama miniature pigs. The microfractures induced mature fibrous repair six months after surgery, while early stages of repair occurred within six weeks. Two processes may occur after microfracture, normal hyaline cartilage regeneration and abnormal fibrocartilage repair. The study identified key cell subsets and their potential roles in these processes, including regulatory chondrocytes, proliferative chondrocytes, cartilage progenitor cells, skeletal stem cells, macrophages, and endothelial cells.
