Extracellular protein homeostasis in neurodegenerative diseases

A study reviews the mechanisms of extracellular proteostasis and its impairment in age-related neurodegenerative diseases. The proteostasis system regulates protein synthesis, folding, trafficking, and degradation. Constitutively secreted extracellular chaperones are emerging as key players in protecting neurons and brain cells from extracellular protein aggregates' toxicity and promoting their safe clearance and disposal. The study examines the latest understanding of key molecules and processes that protect the brain from the pathological consequences of extracellular protein aggregation and proteotoxicity. Finally, based on this emerging knowledge, the study contemplates possible therapeutic opportunities for neurodegenerative diseases.
