This study evaluated the safety, tolerability, and pharmacodynamics of GLY-200, an orally administered therapeutic polymer, as a noninvasive alternative for managing type 2 diabetes (T2D) and obesity. The polymer was designed to enhance the barrier function of mucus in the gastrointestinal tract, mimicking the effects of metabolic surgery or duodenal exclusion devices. The results showed no safety concerns, with mild to moderate gastrointestinal events being the most common side effects. Pharmacodynamic effects, such as reductions in glucose and insulin levels and increases in bile acids and gut hormones, were observed in subjects receiving GLY-200 compared to the placebo group. These findings suggest that GLY-200 has the potential to achieve duodenal exclusion noninvasively and support further development for the treatment of T2D and obesity.
