The AIEOP-BFM ALL 2009 study examined the impact of intensified pegylated L-asparaginase (PEG-ASNASE) on high-risk ALL patients during the consolidation phase. Among 1,097 patients meeting high-risk criteria, 809 were randomly assigned to receive or not receive PEG-ASNASE. The analysis revealed no significant decrease in minimal residual disease (MRD) or improvement in outcomes with PEG-ASNASE. Event-free survival at 5 years was comparable, but adverse reactions were higher with PEG-ASNASE. The study suggests challenges in achieving additional benefits with conventional chemotherapy in intensive protocols.
