Functional α7 nicotinic receptors in human airway smooth muscle increase intracellular calcium concentration and contractility in asthmatics

Recent research has revealed that alpha7 nicotinic acetylcholine receptors (α7nAChRs) are expressed in human airway smooth muscle (hASM), including in individuals with asthma. The functionality of these receptors in hASM cells was investigated in both normal individuals and those with mild-moderate asthma. The study found that α7nAChRs were present in the plasma membrane of hASM cells, with higher levels observed in asthmatic individuals. Patch clamp recordings indicated the presence of functional α7nAChR channels, and real-time fluorescence imaging showed that nicotine, acting via α7nAChRs, increased intracellular calcium levels independently of acetylcholine effects, particularly in asthmatic hASM. Additionally, nicotine-induced contractility was observed in hASM, including in individuals with asthma. These findings suggest that α7nAChRs could be a potential target for mitigating airway hyperresponsiveness in asthma.
