High-dose, short-duration versus standard rifampicin for tuberculosis preventive treatment

A recent study compared different doses and durations of rifampicin for Tuberculosis Preventive Treatment (TPT). Among 1368 participants, completion rates were highest in the standard-dose group (82%), followed by the 20 mg/kg group (71%) and the 30 mg/kg group (65%). Grade 3 hepatotoxicity was rare but slightly higher in the 30 mg/kg group. The risk of adverse events was also higher in the 30 mg/kg group compared to the standard and 20 mg/kg groups. The study concludes that 30 mg/kg rifampicin isn't recommended for TPT due to safety and completion concerns, while 20 mg/kg shows similar safety but lower completion rates compared to the standard dose.
