
In a significant development, the study unveiled a cutting-edge natural killer (NK) cell differentiation system using human pluripotent stem cells (hPSCs). This breakthrough technology allows for the genetic modification of NK cells during pluripotency, paving the way for the production of modified "off-the-shelf" hPSC-NKs. The study successfully demonstrated the engraftment potential of hematopoietic progenitor cells (HPCs) and the potent cytotoxicity of hPSC-NKs against various cancers, including leukemia, lymphoma, diffuse intrinsic pontine glioma (DIPG), and glioblastoma (GBM). These modified NK cells also exhibited advantages in terms of checkpoint inhibitor and metabolic gene expression.
