The article describes a phase II clinical trial investigating the effect of ixazomib, a new generation oral proteasome inhibitor, on bone formation and microstructure in patients with multiple myeloma (MM). The results show that ixazomib decreases bone resorption and promotes longer bone formation events, resulting in increased bone volume per total volume. The treatment did not change osteoclast number or COLL1A1High-expressing osteoblasts on bone surfaces, but significantly more superficial bone structural units were enlarged after treatment. These findings suggest that ixazomib has the potential to be a valuable candidate for future maintenance treatment in MM.
