Inhibiting the Deubiquitinase UCHL1 Reduces SARS-CoV-2 Viral Uptake by ACE2

This study examined the potential of ACE2 (angiotensin-converting enzyme 2) as a therapeutic target for COVID-19 caused by SARS-CoV-2. The researchers identified a mechanism for ACE2 protein modulation by the deubiquitinase enzyme UCHL1 and found that overexpression of UCHL1 preserved ACE2 protein abundance while silencing UCHL1 destabilized ACE2. A small molecule inhibitor of UCHL1 DUB activity decreased ACE2 protein concentrations and inhibited SARS-CoV-2 infection in epithelial cells, suggesting that UCHL1 could be a potential therapeutic target to modulate COVID-19 viral entry.
