Inhibition of the Cellular Deubiquitinase UCHL1 Suppresses SARS-CoV-2 Replication

A study has made significant strides in understanding the role of host factors in COVID-19 severity. The analysis discovered that UCHL1 (ubiquitin C-terminal hydrolase 1), a cellular enzyme, plays a pivotal role in SARS-CoV-2 infection. In an in vitro model of human airway epithelium, the study found that inhibiting UCHL1 with LDN-57444 reduced SARS-CoV-2 replication, including various variants, by up to 99.2%. Importantly, this antiviral effect is independent of viral proteases. UCHL1 modulation may provide a novel avenue for COVID-19 treatment and a potential biomarker for susceptibility.
