
The study identified Sine oculis homeobox homolog 1 (SIX1) as a novel driver of prostate cancer (PC) and explored its regulatory mechanisms. Ku70 was found to interact with SIX1 in the nucleus robustly, forming a complex that enhances SIX1-mediated transcriptional activation of glutamate-pyruvate transaminase 2 (GPT2). This interaction promotes the production of alpha-ketoglutarate (α-KG) and is crucial for PC cell proliferation and migration. Notably, GPT2 expression correlated with Ku70 and SIX1 levels in PC tissues, underscoring the importance of the Ku70-SIX1-GPT2 axis in prostate cancer development.
