In a retrospective analysis of 3266 patients with borderline resectable oral cavity squamous cell carcinoma, a 3-drug neoadjuvant chemotherapy (NACT) regimen (Docetaxel-Cisplatin-5-FU) significantly improved overall survival (OS) over a 2-drug regimen. Of the patients, 32.9% received more than two drugs in NACT, with an overall response rate of 32.5%. Those who underwent surgery after NACT had a 10-year OS of 21.8%, compared to 4.1% for those who did not. Patients achieving pathological complete response demonstrated a 5-year OS of 45.3%. The study suggests NACT's substantial role in long-term survival benefits for borderline resectable oral cavity cancer.
