MAPK-negative feedback regulation confers dependence to JAK2-V617F signaling

The study reveals a potential breakthrough in treating JAK2V617F-driven myeloproliferative neoplasms (MPNs). Scientists have discovered that the convergence of cytokine signaling and JAK2V617F mutation induces the MAPK negative regulator, DUSP1. They lead to downregulating p53 and a higher apoptotic threshold, resulting in treatment resistance. However, inhibiting DUSP1 using a small molecule inhibitor and restoring clonal selectivity through Dusp6 effectively eradicated JAK2V617F cells. Targeting DUSP1 selectively could potentially offer a curative response for individuals with JAK2V617F-driven MPNs.
