Researchers developed a model to study phenotypic and functional reprogramming of Schwann cells (SCs) in the skin adjacent to melanoma tumors. This study shows that SCs in the skin adjacent to melanoma tumors are reprogrammed into repair-like immunosuppressive cells with dysregulated lipid oxidation. These cells downregulate pro-inflammatory genes and pathways and produce anti-inflammatory metabolites that suppress anti-tumor T-cell activation. The findings suggest melanoma-associated peritumoral SCs modulate locoregional and systemic anti-tumor immune responses.
