
A study unveiled the effects of metformin in countering nutrient overstimulation's impact on insulin production. Isolated human pancreatic islets were subjected to high glucose and palmitate levels for varying durations, with metformin introduced after 12 hours. The results were striking. For human islets, proinsulin and insulin levels surged initially but diverged after 72 hours, with proinsulin rising while insulin plateaued. However, metformin administration normalized both insulin forms and restored the expression of critical enzymes. In a separate investigation involving adolescents with obesity and diabetes, metformin treatment reduced fasting proinsulin and insulin concentrations while improving glycaemic control. Proinsulin/insulin ratios were enhanced, promising news for diabetes management.
