MiR-100-5p/miR-125b-5p Mimics Suppress Germ Cell Tumor Growth

Combination treatment with miR-100-5p/miR-125b-5p mimics in malignant germ cell tumors (GCTs) significantly inhibited cell growth and disrupted cell cycle progression in seminoma and yolk sac tumor cells. Quantitative RT-PCR and pyrosequencing revealed downregulation and hypermethylation of miR-100-5p/miR-125b-5p loci on chromosomes 11 and 21. 5-azacytidine treatment upregulated these miRNAs, suggesting epigenetic silencing. miRNA mimic replenishment reduced LIN28A levels and increased let-7 tumor suppressor miRNAs, fostering a sustained inhibitory effect on cell phenotype. Knockdown of TRIM71, a miRNA target, mirrored the effects of miRNA replenishment. These results indicate that miR-100-5p/miR-125b-5p mimics or TRIM71 targeting may offer new therapeutic strategies, with further studies on combinations with platinum-based chemotherapy warranted.
