Monitoring of circulating tumor DNA in advanced pancreatic ductal adenocarcinoma predicts clinical outcome and reveals disease progression earlier than radiological imaging

A prospective study investigated the role of longitudinal circulating tumor DNA (ctDNA) measurements in advanced pancreatic ductal adenocarcinoma (PDAC) patients undergoing palliative chemotherapy. Using ctDNA analysis, researchers found that ctDNA detection at baseline independently predicted progression-free and overall survival. Longitudinal ctDNA measurements during chemotherapy successfully detected disease progression earlier than radiological imaging, providing a median lead time of 23 days. The study confirmed the clinical relevance of ctDNA in advanced PDAC, demonstrating its value in predicting clinical outcomes and monitoring disease progression during treatment.
