
The phase III CheckMate 8HW trial confirmed the efficacy of nivolumab plus ipilimumab as first-line therapy for unresectable or metastatic mismatch repair deficient (dMMR) or microsatellite instability-high (MSI-H) colorectal cancer (CRC). Among 303 patients randomized (2:1) to receive nivolumab plus ipilimumab or investigator's choice chemotherapy, the primary endpoint was progression-free survival (PFS) in 255 patients with centrally confirmed dMMR/MSI-H CRC. This study builds on the promising results of the earlier phase II CheckMate 142 trial, supporting the combination's role as a standard first-line treatment in this patient population.
