
This study focuses on optimizing skin surface metabolomics by evaluating various sampling materials, extraction solvents, taping methods, and analytical techniques. The findings indicate that using D-Squame® D100 tape with a methyl tert-butyl ether/methanol extractant is ideal for lipid analysis. A five-time taping procedure with a single tape provides sufficient biomass for lipidomics, while different methods are necessary for water-soluble compounds. Additionally, associations among skin surface metabolites were identified, shedding light on microbial cutotypes and host-microbe interactions.
