
Osimertinib induces apoptosis in EGFR mutant (EGFRm) NSCLC cells through TRAIL (TNFSF10) upregulation, which is mediated by IL6/STAT3 signaling. This study reveals that osimertinib activates STAT3 to increase TRAIL expression and enhances DR5 clustering, crucial for apoptosis. The findings highlight TRAIL’s role in osimertinib efficacy and resistance mechanisms, presenting new insights into targeting the IL6/STAT3/TRAIL axis to improve NSCLC treatment outcomes.
