Palmitic acid induces GSDMD‐mediated pyroptosis in periodontal ligament cells via the NF‐κB pathway

This study investigated the effects of palmitic acid (PA), a saturated fatty acid elevated in obese individuals, on pyroptosis (inflammatory cell death) in periodontal ligament cells (PDLCs) and its underlying mechanisms. The researchers isolated human PDLCs and exposed them to PA. They observed pyroptotic characteristics in PDLCs treated with PA, including cell swelling, membrane rupture, and changes in organelles. PA also induced inflammatory responses and increased IL-1β levels. The study identified caspase-1, caspase-4, and GSDMD as proteins involved in PA-induced cell death. Additionally, PA activated the NF-κB pathway, and inhibiting NF-κB partially rescued cell death caused by PA.
