Pancreatic ductal adenocarcinoma induces neural injury that promotes a transcriptomic and functional repair signature by peripheral neuroglia

New insights into the understanding of perineural invasion (PNI) in pancreatic ductal adenocarcinoma (PDAC) have been uncovered through a study utilizing digital spatial profiling. The study identified changes in the transcriptome and neural-supportive cell types within the tumor-nerve microenvironment of PDAC during PNI. Hypertrophic tumor-associated nerves were found to express signals of nerve damage, including programmed cell death and Schwann cell proliferation signaling pathways. The study also revealed increased local neuroglial cell proliferation and frequent apoptotic cell turnover. Functional calcium imaging studies confirmed neuronal activity and apoptosis in nerve bundles.
