A recent double-blind, randomized clinical trial conducted across 43 centres in 5 countries aimed to evaluate the efficacy of pegargiminase-based chemotherapy on survival in patients with nonepithelioid pleural mesothelioma, an arginine-auxotrophic tumour. Participants received either pegargiminase or a placebo alongside standard chemotherapy. Results showed extended overall survival (9.3 vs 7.7 months) and progression-free survival (6.2 vs 5.6 months) with pegargiminase. Adverse events were manageable. These findings suggest pegargiminase-based chemotherapy could offer improved outcomes for this challenging cancer type, warranting further clinical investigation.
