
Hematopoietic stem cells (HSCs) play a crucial role in maintaining blood cell production throughout an organism's life. However, disruptions in their regulation can lead to blood cancers. This study have found that a protein called PPM1D, which negatively regulates the tumor suppressor p53, influences HSC behavior. Truncated PPM1D, resulting from mutations, reduces HSC self-renewal under normal conditions but promotes aggressive acute myeloid leukemia (AML) after exposure to radiation. Inhibiting PPM1D curbs the growth of leukemic cells with truncated PPM1D, offering protection against radiation-induced cell growth. This discovery suggests potential therapeutic avenues for AML treatment.
