
This study investigated factors influencing malignant transformation (MT) in localized oral leukoplakia (OL) and proliferative verrucous leukoplakia (PVL). Data on demographics, clinical features, histology, and DNA ploidy were collected from 133 OL and 20 PVL patients over 6 years. Oral epithelial dysplasia (OED), DNA ploidy, clinical presentation, and lesion site correlated with MT in OL patients in univariate analysis. OED emerged as the strongest MT predictor in multivariate modeling, with DNA ploidy enhancing prediction. In PVL patients, no predictors related to MT were identified. DNA ploidy might aid in identifying low-risk OL subsets for MT.
