
A study investigated the mechanism behind bone regeneration in distraction osteogenesis (DO). Using canine models, they compared DO with bone defects (BD) and found that DO resulted in increased bone formation and mineralization, while BD showed incomplete bone union. They isolated and identified mesenchymal stem cells (MSCs) from DO and BD calluses, discovering that DO-MSCs exhibited stronger osteogenic capabilities. Single-cell RNA sequencing analysis revealed distinct cell populations, including PRRX1+MSCs associated with epithelial-mesenchymal transition. Knocking out PRRX1 in DO hindered jawbone regeneration, indicating its crucial role. This study provides valuable insights into DO regeneration and the importance of PRRX1+MSCs.
