Reversible Male Contraception through ALDH1A Inhibition in Mice

Recent research has shed light on the effects of ALDH1A1/1A2 inhibition on male fertility, focusing on the potential for WIN 18,446 as a male contraceptive. The study found that a 4-week treatment with WIN 18,446 in male mice significantly disrupted spermatogenesis, leading to a progressive decline in testes weight and seminiferous epithelium damage. However, the inhibition was reversible, with fertility fully restored after 9 weeks of recovery. The findings suggest that strong, episodic inhibition of ALDH1A may offer a novel, reversible approach to male contraception by blocking spermatogenesis without permanent damage.
