This study investigated the effectiveness of single-dose rifapentine in preventing leprosy in household contacts of leprosy patients. The trial involved three groups: rifapentine, rifampin, and no intervention (control). The primary outcome was the cumulative incidence of leprosy over a 4-year period. The results showed that the cumulative incidence was 0.09% with rifapentine, 0.33% with rifampin, and 0.55% without intervention. The rifapentine group had an 84% lower incidence than the control group. In contrast, there was no significant difference between the rifampin group and the control group. No severe adverse events were observed. Therefore, single-dose rifapentine demonstrated a lower incidence of leprosy compared to no intervention.
