
This study, conducted within the Rotterdam Study, explores the association between skin advanced glycation end products (AGEs) measured as skin autofluorescence and the risk of dementia. AGEs are compounds formed due to conditions like hyperglycemia and oxidative stress implicated in dementia. Skin autofluorescence was measured in 2922 participants without dementia, and 1504 also underwent brain magnetic resonance imaging (MRI) to assess brain atrophy and cerebral small vessel disease. The participants were followed until 2020 for dementia incidence. Higher skin autofluorescence was independently associated with an increased risk of dementia and Alzheimer’s disease. The study suggests a potential involvement of AGEs in dementia pathophysiology.
