Sorafenib maintenance after allogeneic haemopoietic stem-cell transplantation in patients with FLT3-ITD acute myeloid leukaemia: long-term follow-up of an open-label, multicentre, randomised, phase 3 trial

Sorafenib maintenance after stem cell transplantation improves survival and reduces relapse in patients with FLT3 internal tandem duplication (FLT3-ITD) acute myeloid leukemia, according to a phase 3 trial conducted in China. The trial involved 202 patients who were randomly assigned to receive sorafenib maintenance or no maintenance after transplantation. The 5-year follow-up data showed that the sorafenib group had higher overall survival, leukemia-free survival, and graft-versus-host disease-free, relapse-free survival compared to the control group. The cumulative incidence of relapse was lower in the sorafenib group, with no increase in non-relapse mortality. The findings support sorafenib maintenance as a standard treatment for FLT3-ITD acute myeloid leukemia patients undergoing transplantation.
