Superiority of low dose benzbromarone add-on to low dose febuxostat compared to febuxostat monotherapy in gout

This prospective, randomized trial addressed the unmet need for simplified urate-lowering therapy (ULT) regimens in gout patients with combined renal urate underexcretion and overload. The study compared febuxostat monotherapy with a combination of febuxostat and low-dose benzbromarone. Among 250 participants, the febuxostat and benzbromarone group exhibited a significantly higher proportion of achieving the serum urate target (<360μmol/L) at 12 weeks compared to febuxostat monotherapy (75.5% vs 47.7%). Safety profiles were comparable. Adding low-dose benzbromarone to low-dose febuxostat demonstrated superior urate lowering, providing a potential alternative with expedited target achievement for selected gout patients.
